    La dynamique de la Technologie Intranet au service d'un système d'information avec Bases de données interactives.

    International audienceInternet s'accroît de manière exponentielle depuis le début des années 90. Ce phénomène est dû en partie aux entreprises qui ont vu dans cette technologie l'outil par excellence pour élargir leur marché et étudier leur environnement. L'information est aujourd'hui au coeur de la vie de l'entreprise. Mais elle est aussi une matière première indispensable qui est souvent désorganisée aussi bien dans sa collecte que dans son traitement et sa diffusion. La solution pour délivrer l'information " just-in-time " est l'implémentation d'un système de communication universel. Un réseau qui relierait tous les intervenants de l'entreprise s'apparente manifestement à l'Internet. Et, utiliser Internet au sein des entreprises, c'est le principe des Intranets. En effet, l'Intranet permet une communication sécurisée et transversale des "intranautes", ainsi qu'un apport en intelligence de l'entreprise. La gestion optimale de cette plus-value passe par une interactivité de l'Intranet. Pour ce faire, un système de Bases de Données doit être construit. Ces bases peuvent concerner l'information interne à l'entreprise et sa mémoire (compte-rendu des congrès/colloque, annuaires, ...) ainsi que l'information externe enrichie par les experts (benchmarking : sites des concurrents, brevets, ...) Cette technologie peut se baser sur plusieurs types d'outils qui se déclinent en suites logicielles ou logiciels intégrés. C'est à l'entreprise de choisir la technologie adéquate en fonction de ses besoins, moyens, et compétences. Pour illustrer cet état de fait, FAURECIA développe un système d'information qui repose sur un Intranet interactif. Pour être de plus en plus réactive sur ses marchés, l'entreprise doit être de plus en plus communicante, en son sein et avec ses partenaires (clientèle et fournisseurs). Pour s'en rapprocher, elle pourra leur ouvrir son Intranet. C'est le concept de l'Extranet. Cette technologie intègre la notion de "l'entreprise étendue", ou "l'entreprise virtuelle". Et parmi les fondements de ce nouveau concept managérial, l'extension du système d'information en amont et aval de l'entreprise.     Large-scale analysis by SAGE reveals new mechanisms of v-erbA oncogene action

    <p>Abstract</p> <p>Background:</p> <p>The <it>v-erbA </it>oncogene, carried by the Avian Erythroblastosis Virus, derives from the <it>c-erbAα </it>proto-oncogene that encodes the nuclear receptor for triiodothyronine (T3R). v-ErbA transforms erythroid progenitors <it>in vitro </it>by blocking their differentiation, supposedly by interference with T3R and RAR (Retinoic Acid Receptor). However, v-ErbA target genes involved in its transforming activity still remain to be identified.</p> <p>Results:</p> <p>By using Serial Analysis of Gene Expression (SAGE), we identified 110 genes deregulated by v-ErbA and potentially implicated in the transformation process. Bioinformatic analysis of promoter sequence and transcriptional assays point out a potential role of c-Myb in the v-ErbA effect. Furthermore, grouping of newly identified target genes by function revealed both expected (chromatin/transcription) and unexpected (protein metabolism) functions potentially deregulated by v-ErbA. We then focused our study on 15 of the new v-ErbA target genes and demonstrated by real time PCR that in majority their expression was activated neither by T3, nor RA, nor during differentiation. This was unexpected based upon the previously known role of v-ErbA.</p> <p>Conclusion:</p> <p>This paper suggests the involvement of a wealth of new unanticipated mechanisms of v-ErbA action.</p

    Combined transcriptome studies identify AFF3 as a mediator of the oncogenic effects of β-catenin in adrenocortical carcinoma

    International audienceAdrenocortical cancer (ACC) is a very aggressive tumor, and genomics studies demonstrate that the most frequent alterations of driver genes in these cancers activate the Wnt/β-catenin signaling pathway. However, the adrenal-specific targets of oncogenic β-catenin-mediating tumorigenesis have not being established. A combined transcriptomic analysis from two series of human tumors and the human ACC cell line H295R harboring a spontaneous β-catenin activating mutation was done to identify the Wnt/β-catenin targets. Seven genes were consistently identified in the three studies. Among these genes, we found that AFF3 mediates the oncogenic effects of β-catenin in ACC. The Wnt response element site located at nucleotide position − 1408 of the AFF3 transcriptional start sites (TSS) mediates the regulation by the Wnt/β-catenin signaling pathway. AFF3 silencing decreases cell proliferation and increases apoptosis in the ACC cell line H295R. AFF3 is located in nuclear speckles, which play an important role in RNA splicing. AFF3 overexpression in adrenocortical cells interferes with the organization and/or biogenesis of these nuclear speckles and alters the distribution of CDK9 and cyclin T1 such that they accumulate at the sites of AFF3/speckles.     FOXL2 is a Progesterone Target Gene in the Endometrium of Ruminants

    Forkhead Box L2 (FOXL2) is a member of the FOXL class of transcription factors, which are essential for ovarian differentiation and function. In the endometrium, FOXL2 is also thought to be important in cattle; however, it is not clear how its expression is regulated. The maternal recognition of pregnancy signal in cattle, interferon-Tau, does not regulate FOXL2 expression. Therefore, in the present study, we examined whether the ovarian steroid hormones that orchestrate implantation regulate FOXL2 gene expression in ruminants. In sheep, we confirmed that FOXL2 mRNA and protein was expressed in the endometrium across the oestrous cycle (day 4 to day 15 post-oestrus). Similar to the bovine endometrium, ovine FOXL2 endometrial expression was low during the luteal phase of the oestrous cycle (4 to 12 days post-oestrus) and at implantation (15 days post-oestrus) while mRNA and protein expression significantly increased during the luteolytic phase (day 15 post-oestrus in cycle). In pregnant ewes, inhibition of progesterone production by trilostane during the day 5 to 16 period prevented the rise in progesterone concentrations and led to a significant increase of FOXL2 expression in caruncles compared with the control group (1.4-fold, p < 0.05). Ovariectomized ewes or cows that were supplemented with exogenous progesterone for 12 days or 6 days, respectively, had lower endometrial FOXL2 expression compared with control ovariectomized females (sheep, mRNA, 1.8-fold; protein, 2.4-fold; cattle; mRNA, 2.2-fold; p < 0.05). Exogenous oestradiol treatments for 12 days in sheep or 2 days in cattle did not affect FOXL2 endometrial expression compared with control ovariectomized females, except at the protein level in both endometrial areas in the sheep. Moreover, treating bovine endometrial explants with exogenous progesterone for 48h reduced FOXL2 expression. Using in vitro assays with COS7 cells we also demonstrated that progesterone regulates the FOXL2 promoter activity through the progesterone receptor. Collectively, our findings imply that endometrial FOXL2 is, as a direct target of progesterone, involved in early pregnancy and implantation

These attributes have made these species models for understanding the complexity of necrotrophic, broad host-range pathogenicity. Despite their similarities, the two species differ in mating behaviour and the ability to produce asexual spores. We have sequenced the genomes of one strain of S. sclerotiorum and two strains of B. cinerea. The comparative analysis of these genomes relative to one another and to other sequenced fungal genomes is provided here. Their 38–39 Mb genomes include 11,860–14,270 predicted genes, which share 83% amino acid identity on average between the two species. We have mapped the S. sclerotiorum assembly to 16 chromosomes and found large-scale co-linearity with the B. cinerea genomes.     Sensitivity of global ocean biogeochemical dynamics to ecosystem structure in a future climate

    Terrestrial and oceanic ecosystem components of the Earth System models (ESMs) are key to predict the future behavior of the global carbon cycle. Ocean ecosystem models represent low complexity compared to terrestrial ecosystem models. In this study we use two ocean biogeochemical models based on the explicit representation of multiple planktonic functional types. We impose to the models the same future physical perturbation and compare the response of ecosystem dynamics, export production (EP) and ocean carbon uptake (OCU) to the same physical changes. Models comparison shows that: (1) EP changes directly translate into changes of OCU on decadal time scale, (2) the representation of ecosystem structure plays a pivotal role at linking OCU and EP, (3) OCU is highly sensitive to representation of ecosystem in the Equatorial Pacific and Southern Oceans

Their 38–39 Mb genomes include 11,860–14,270 predicted genes, which share 83% amino acid identity on average between the two species. We have mapped the S. sclerotiorum assembly to 16 chromosomes and found large-scale co-linearity with the B. cinerea genomes. Seven percent of the S. sclerotiorum genome comprises transposable elements compared to <1% of B. cinerea. The arsenal of genes associated with necrotrophic processes is similar between the species, including genes involved in plant cell wall degradation and oxalic acid production. Analysis of secondary metabolism gene clusters revealed an expansion in number and diversity of B. cinerea–specific secondary metabolites relative to S. sclerotiorum. The potential diversity in secondary metabolism might be involved in adaptation to specific ecological niches. Comparative genome analysis revealed the basis of differing sexual mating compatibility systems between S. sclerotiorum and B. cinerea. The organization of the mating-type loci differs, and their structures provide evidence for the evolution of heterothallism from homothallism. These data shed light on the evolutionary and mechanistic bases of the genetically complex traits of necrotrophic pathogenicity and sexual mating. This resource should facilitate the functional studies designed to better understand what makes these fungi such successful and persistent pathogens of agronomic crops
